I can record a strong signal with my Strat on a mono track with GTR Tuner as the only Insert, but there is no signal in the tuner plugin. Is there something I am missing?

Did you enable Input Monitoring in Pro Tools to listen to the signal after it has been processed ?
If you still experience issues, I suggest consulting with our Technical Support team, they will gladly assist with solving this and can also remote in to your computer if it is not solved over the email.
